Noopept is a cognitive-enhancing peptide synthesized in Russia in 1996, as a successor to prototypical nootropic drug Piracetam. It was based off the endogenous neuropeptide cycloprolylglycine. Peptides are molecules consisting of 2 or more amino acids linked together by peptide bonds. Depending on the number of amino acids, peptides are called dipeptides, tripeptides, tetrapeptides, etc.
As said before, Noopept is a dipeptide. Peptides are not well absorbed in the GI tract and need to be injected via intramuscular or intravenous injection. Noopept, however, is a peptide that is only two amino acids long (dipeptide), and this allows it to be small enough to be absorbed orally, without being ripped apart by digestive enzymes and acids.
It is commonly thought to be a racetam, but it is not since it does not have a pyrrolidinone nucleus.
It is said to be 1000 times stronger than Piracetam in effective dose level and nootropic activity, however, this statement doesn’t seem to have a scientific foundation.
The advantage of Noopept over Piracetam is its ability to stimulate the expression of the neurotropic factors NGF and BDNF.
NGF(Nerve Growth Factor) prevents neuronal degeneration and promotes myelin repair. Dysregulation of NGF signaling has also been linked to Alzheimer’s disease and other psychiatric disorders, such as dementia, depression, schizophrenia, autism.
BDNF(Brain-derived neurotrophic factor), like NGF, plays a significant role in neurogenesis. It controls synaptic function and plasticity and modulates neuronal survival.
The major metabolite of Noopept, cycloprolylglycine, has anxiolytic effect in animal models. Scientists hypothesize this effect is a consequence of increased inhibitory transmission in the hippocampus.
Another interesting effect of Noopept is its immune boosting ability. It reduces immunosuppression induced by cyclophosphamide, stimulates immune response to various antigens, and increases phagocytic activity of macrophages.

How to Take
Noopept is sold in Russia in 10 mg tablets to be taken 2-3 times a day, but the most common dose is 20 mg twice a day.
Even though it is orally bioavailable, anecdotal reports indicate that oral and sublingual routes of absorption have different effects. The sublingual route has a stronger nootropic effect while the oral route is slightly more anxiolytic. No human studies comparing the absorption of Noopept via different routes have been conducted, however.

The effect of Noopept becomes stronger with chronic treatment and the official drug package insert recommends taking Noopept in a “3 months on, 1 month off” cycle.
Noopept doesn’t have serious side effects or contraindications.
The most common reported side effects are irritability and problems with short-term memory. These side effects usually go away after a week of dosing and can be a consequence of excessively high dose.
Most nootropics have a bell-shaped response curve, so taking a higher dose than recommended will not necessarily make the positive effects stronger and could potentially increase the negative effects.

Piracetam is considered by many to be the father of all nootropic drugs. It has a history of being used to treat dementia, Alzheimer’s Disease and other cognitive diseases that come with old age. Most research conducted on piracetam has come to the consensus that it does not have much effect on individuals who are not experiencing cognitive decline. For this reason, piracetam does not seem like an ideal alternative treatment for attention deficits. However, piracetam is extremely safe to take, and supplementation by anyone will help prevent cognitive decline before it even starts. Thus, it might be worthwhile to take piracetam alongside another medication on this list. One small study mentions a combination of atomoxetine (an ADHD medication) and Piracetam.

It is therefore frequently combined with stimulants to reduce their neurotoxic effects as well as reducing tolerance to the positive effects of Adderall and Ritalin. Some research has been done on memantine’s possible effectiveness in treating ADHD (by itself, not in combination). One study found that memantine was fairly beneficial for alleviating symptoms of ADHD, but concluded that there is not enough evidence to draw any real conclusions. Modafinil is a wakefulness-promoting (eugeroic) drug that is classified in many places as a prescription medication. Armodafinil, a closely related drug, consists of only the active (−)-(R)-enantiomer of modafinil, meaning it is theoretically more potent. Because of this, both drugs work in a similar fashion. Modafinil has shown a good amount of promise as an alternative treatment of ADD/ADHD. One study conducted on children found that 48% of the participants felt a significant improvement in attentive skills while on modafinil. Although modafinil’s mechanism works through modulation of dopamine, it does not seem to carry an addiction potential to the same degree as amphetamines.
Selegiline (L-deprenyl) is a substituted phenethylamine drug commonly used to treat Parkinson’s disease and dementia. It has also seen some use as an alternative treatment for depression. There has not been a significant amount of research done on its efficacy in treating ADHD, but one study done on a small group of 28 children with ADHD studied the treatment effects of selegiline in comparison to methylphenidate. The children treated with selegiline displayed fewer symptoms of ADHD than those treated with methylphenidate while also displaying fewer side effects. This research is preliminary, but it demonstrates the selegiline displays promise as an ADHD treatment.

For some, engulfing various powders every morning is a tedious task but others may embrace it as part of their daily ritual. However, you may not be getting the most out of what you’re consuming due to the metabolic breakdown in the liver and the contents lost to the digestive system. Often in the nootropics community we forget about other, more efficient methods of administration in lieu of the easiest route. They all come with their own benefits and downfalls, and certain methods are optimal for certain drugs.
Like many of us, for a long time I was content to place powder in my mouth and swallow. I was unaware that there were far more productive ways to achieve the cognitive enhancement I desired. Prolonged periods of research lead me to uncover the benefits of sublingual administration.  Whilst the effects of Noopept became more potent, a misplaced particle was enough to send me gagging.
Many nootropic substances have a low bioavailability, which leads to oral administration being less effective. Eventually I stumbled upon intranasal administration, a technique that yielded superior results to those I had previously encountered.
A pivotal concept in the usage of nootropics is to pass the brain barrier. Nasal administration allowed me to perfect opportunity to do so. The cavity of the nose is covered in thin mucosa which are well vascularised making it an ideal entry point for nootropics.  The nose enables certain drugs and other molecules to bypass the blood brain barrier via diffusion through the olfactory epithelium and the perineural sheath or via retrograde axonal transport along olfactory and trigeminal nerves. Certain nootropic agents like Adrafinil shouldn’t be absorbed via the nose due to the active ingredients that need to be processed by the liver. 
After browsing many web pages, online forums and blogs, I found it was easy to procure pre-made nasal sprays capable of dispensing nootropics. However, I’d decided to create my own mixture purely out of curiosity more so than economic benefit. I purchased a 10 ml nasal spray bottle that would be capable of delivering a 4 mg spray of Noopept.
I measured out 400 mg of Noopept then added 10ml of Bacteriostatic water and begun to dissolve the particles by syphoning in and out of a blunt syringe. Noopept was particularly stubborn in breaking down its particles, but after 10 minutes the solution was dissolved to my satisfaction. Soon after mixing, it didn’t take long for me to administer my first dosage. Placing the end of the bottle at my nostril, I pressed the trigger twice for a total dose of 8 mg.

I felt the effects within 15 minutes of application. It was at this point I realised that nasal administration increased the onset of Noopept. This is due to some smaller molecules being absorbed in as little as 5 minutes through the nose. 
The effects were not subtle like my earlier experiences with oral Noopept.
An immediate clarity settled over my head. Music seemed more vivid with a sudden depth to every beat. I was now in control of my every thought and emotion. The muddled parts of my brain were able to be organised into coherent thinking, which was easily verbalised without haste in my expression.
My social anxiety was subdued, which lead to my concentration to become far more precise and calculated. The influence of Noopept lasted significantly longer than ever before. An 8 am dosage remained noticeable until 4 pm. A miraculous increase in the observable effects compared to my previous trials with oral Noopept.
The positive outcome of this experiment encouraged me to continue dosing for another 4 weeks where the results remained consistently pleasing. My daily motivation for activity was greater than ever before and I was invigorated in the completion of even menial tasks.
Overall, there was a noticeable increase in my brain function. I could never claim it increased my IQ. But it certainly unlocked my ability to use my intellect far more effectively. At the end of each month, I initiated a short break to refresh my brain chemistry and reduce any tolerance that had grown, and then resumed my supplementation a few weeks later.
